Gly-Gly-Leu is usually combined with a self-immolative spacer (e.g., PABC: para-aminobenzyloxycarbonyl).
General structure:
Antibody – Linker (Gly-Gly-Leu-PABC) – Drug
After enzymatic cleavage, the spacer undergoes self-immolation, releasing the free payload.
